摘    要:目的:探索脂肪成体干细胞中是否表达胚胎时期标志蛋白SSEA-1,OCT-4,GRB.方法:先将C57小鼠脂肪用I型胶原酶消化,然后采用直接贴壁法,培养获得的细胞.取第3代生长状态稳定、形态均一的细胞进行成脂、成骨诱导分化,同时观察所培养的第3代细胞表达胚胎时期标志蛋白SSEA-1,OCT-4,GRB的情况.结果:通过该培养方法,可以获得稳定的细胞群体,并能成功进行成骨、成脂诱导分化;免疫荧光化学检测显示三种胚胎时期出现的特殊蛋白均有不同程度的表达.结论:脂肪来源的成体干细胞中表达胚胎时期的产物,这种特性可能与干细胞的多向分化特性有关系.

Abstract:AIM:To observe the embryo symbol proteins(SSEA-1,OCT-4 and GRB)in mouse adipose-derived adult stem cells(ASCs).METHODS:ASCs were obtained from C57 mouse adipose tissues and were cultured in DMEM/F12 with 100 mL/L fetal bovine serum.The third passage of ASCs was induced to differentiate after exposure to adipocytic or osteogenic medium.The expressions of proteins(SSEA-1,OCT-4 and GRB)at the embryonic stage of the third passage of ASCs were observed.RESULTS:The cells isolated from adipose tissue of C57 mice h...
