光动力疗法影响大鼠C6胶质瘤AQP4表达的实验研究

摘    要:目的研究光动力疗法(photodynamic therapy,PDT)治疗大鼠c6脑胶质瘤后水通道蛋白4(aquaporin4,AQP4)的变化,并探究其与脑水肿之间的关系。。方法培养C6胶质瘤细胞,建立c6大鼠胶质瘤模型,分成PDT组、单纯激光照射组、单纯光敏剂组、空白对照组进行实验,术后24h分别用鼠脑干湿重法及Westernblot进行脑水肿对比和AQP4蛋白定量测定。结果PDT组鼠脑组织含水量大于其他三组,而单纯激光照射组与单纯光敏剂组之间无明显差异,但大于空白对照组。AQP4与脑水肿密切相关。AQP4在PDT组表达明显高于单纯激光照射组和单纯光敏剂组,远高于空白对照组。结论光动力治疗脑胶质瘤可加重脑水肿的发生,AQP4在光动力治疗脑胶质瘤术后表达增高,与脑水肿成正相关。AQP4介导了光动力治疗脑胶质瘤术后脑水肿的加重,因而调控AQP4的表达有可能成为光动力治疗脑胶质瘤的新的探索方向。

Abstract:Objective To study the change of AQP4 after photodynamic therapy for rat C6 glioma and explore its relationship with brain edema. Methods C6 glioma cells were cultured to establish C6 rat glioma model and divided into four groups: the photodynamic therapy group, laser group, photosensitizer group, and blank control group. For each group, the brain water content was determined with wet - dry weighting method and AQP4 protein level with Western blot 24 hours after the photodynamic therapy. Results The photodynamic therapy group indicated much more brain water than the other three groups. There was no statistical differ- ence between the laser and the photosensitizer group, but beth of them showed more brain water than the blank control group. As to AQP4 expression, the laser group and the photosensitizer group was much less powerful than the photedynamic therapy group, but more impressive than the blank control group. Conclusions Treating glioma with photedynamic therapy may aggravate brain edema. AQP4 expression was enhanced in the photedy-namic therapy group, so there~ a positive correlation between AQP4 expression and brain edema caused by photodynamic therapy. We can draw the conclusion that the aggravation of brain edema after glioma operation with photodynamic therapy is mediated by AQP4. Therefore, regulating AQP4 expression may be a new direction for us to explore how to treat glioma with photodynamic therapy.
