逆转录病毒介导的野生型p53基因转入对慢粒多药耐药细胞恶性表型的影响

摘    要:以K562/HHT 细胞为模型,采用重组逆转录病毒载体介导的基因转移技术,将人野生型p53cDNA转入该细胞系,研究外源性野生型p53 基因在白血病多药耐药细胞表达的意义。结果显示,外源性野生型p53 基因抑制细胞增殖,一般形态学观察见细胞明显向成熟型分化。流式细胞仪分析还显示p53 基因诱导K562/HHT细胞死亡且伴有细胞周期G1 期的生长阻滞。结果表明,野生型p53 基因对多药耐药的白血病细胞能部分降低其恶性表型

Abstract:To study the significane of expression of exogenous wild type p53 gene(WP53G) in leukemic multidrug resistance cells, human WP53G cDNA was transferred by a recombinant retroviral vector in to K562/HHT cells. The Results suggest that exogenous WP53G inhibit cell growth and induce cell differentiation. Flow cytometric analysis showed that exogenous WP53G induced K562/HHT cells to die, which was accompanied by growth arrest in G 1 phase of the cell cycle. The conclusion can be drawn that the malignant phenotype can be at least partially reversed by restoring WP53G expression.
