大蒜素对人白血病HL-60细胞生长和凋亡的影响

目的: 探讨大蒜素对人白血病HL-60细胞生长抑制和诱导凋亡作用。方法: MTT比色分析法检测大蒜素对HL-60细胞生长的抑制作用,光镜观察细胞凋亡形态学变化,流式细胞术分析大蒜素对HL-60细胞凋亡的影响。结果: 20mg/L、40mg/L、60mg/L大蒜素均能抑制HL-60细胞生长,其抑制作用呈剂量-效应关系,40mg/L大蒜素能诱导HL-60细胞出现典型凋亡形态学变化。大蒜素诱导HL-60细胞有凋亡现象和阻滞细胞周期于G₂/M期。结论: 大蒜素对人白血病HL-60细胞生长有抑制作用,其抑制作用与诱导HL-60细胞凋亡和阻抑细胞周期有关。

Effect of growth and apoptosis on human leukaemia cells line HL-60 induced by allicin

Objective:To study the inhibitory effects of allicin and its induction of apoptosis on human leukemia cells HL-60.Methods:MTT was used to study the inhibitory effects of allicin on HL-60 cells growth and the typical morphology characters of apoptosis induced by allicin was observed with light microscope,and the apoptosis rate was analysed by flow cytometry(FCM).Results:MTT showed that HL-60 cells were suppressed with dose dependent relationship after exposed to allicin of 20,40,60 mg/L.Allicin induced apoptosis of HL-60 cells and apoptotic cells were supposed to be initiated in some phases of cell cycle by flow cytometry.Conclusions:Allicin can inhibit the growth rates of human leukemia cells HL-60 and induce its apoptosis.
