氢醌致细胞凋亡及其机制的体外实验研究

摘    要:目的了解氢醌(HQ)致细胞凋亡作用及其氧化损伤机制。方法用吖啶橙/溴化乙啶(AO/EB)双标记法观察HQ对中国仓鼠肺成纤维细胞(V79)的凋亡作用;用DCFH-DA法检测不同浓度HQ处理V79细胞0.5 h和1 h后活性氧(ROS)的含量。结果AO/EB双标记法显示氢醌对V79细胞有明显促凋亡作用,并呈浓度反应关系。DCFH-DA法检测结果显示,在设置的4个浓度范围内,活性氧含量随着浓度的增加而增加(P<0.01);染毒0.5 h组各浓度ROS含量均高于染毒1 h组各相应浓度(P<0.01),且呈浓度依赖关系。结论在体外培养条件下,低浓度HQ能致V79细胞凋亡,活性氧产生的高峰在0.5 h,表明HQ引起的细胞损伤是一种氧化损伤。

Abstract:Objective To explore the apoptosis and oxidative damage induced by hydroquinone(HQ)on V79 cells in vitro.Methods Apoptosis was evaluated by AO/EB double labeling method.DCFH-DA assay was used to detect the ROS contents caused by various concentration of HQ in V79 cells.Results As showed in the AO/EB double labeling method,apoptosis in V79 cell could be induced obviously by HQ showing concentration-dependent relationship between HQ and apoptosis.The DCFH-DA assay showed that the ROS contents increased with t...
