缺氧性肺动脉高压大鼠肺iNOS表达的实验研究

摘    要:目的 观察慢性缺氧大鼠肺组织诱生型一氧化氮合酶 (iNOS)表达的变化 ,并探讨iNOS在肺动脉高压发病中的作用。 方法 运用血清学检验及免疫组织化学方法观察血一氧化氮 (NO)、肺组织iNOS蛋白表达的变化。 结果 正常组大鼠iNOS表达呈弱阳性 ,慢性缺氧后肺组织iNOS蛋白表达呈强阳性 ,血一氧化氮水平升高 (P <0 0 1 )。 结论 一氧化氮合酶表达增强及一氧化氮参与慢性缺氧性肺动脉高压发病过程 ,诱生型一氧化氮合酶的表达及一氧化氮的血管舒张作用与氧自由基作用的放大效应可能是肺动脉高压形成的重要因素

Abstract:Objective The aim of the study was to observe the immunohistochcmical changes of induced nitric oxide in lung tissue from chronic hypoxia, and to explore the effects of iNOS on the formation of hypoxic pulmonary hypertension. Methods ImmunohistochemicaI techniques was used to assess the changes of iNOS in the lung tissue. Results Expression of iNOS protein in the lung tissue of hypoxic pulmonary hypertension were markedly elevated, and NO was significantly raised in the plasma after hypoxia (P<0.01). Conclusions iNOS and NO may play an important role in the formation of hypoxic pulmonary hypertension(HPT). The imbalance between vascular relaxing effects and oxygen free radical of NO is of importance in the pathogenesis of HPT
