基于网络药理学探讨五味清浊散加减方治疗功能性腹泻的作用机制研究

摘    要:目的:基于网络药理学方法初探五味清浊散加减方治疗功能性腹泻(FD)的潜在活性化合物及其可能的作用靶点通路。方法:运用TCMSP和ETCM数据库获取和筛选五味清浊散加减方的活性成分,在PubChem中查询其结构信息,再利用Swiss Target Prediction和Super Pred预测药物的作用靶点,运用Drug Bank、Gene Cards、OMIM数据库检索FD作用靶点;选择药物与疾病的交集靶点导入STRING,绘制靶点蛋白-靶点蛋白相互作用网络;利用Metascape对关键靶点进行GO和KEGG富集分析,预测五味清浊散加减方作用机制与FD作用靶点通路。结果:共收集到FD相关靶点371个,五味清浊散加减方的活性成分靶点227个,涉及治疗FD靶点51个;GO和KEGG富集分析发现五味清浊散加减方可能作用于SRC、PTPN11、PIK3R1、AKT1、MAPK1、STAT1、SYK等靶点,并通过免疫相关通路、炎症相关通路、扩张平滑肌、癌症等多条通路达到其治疗FD目的。结论:通过网络药理学证明五味清浊散加减方对FD的改善具有多成分、多靶点、多通路协调治疗疾病的特点,为临床应用五味清浊散加减方提供了理论支持。

To explore the mechanism of Wuwei Qingzhuo Powder in the treatment of functional diarrhea based on network pharmacology

Abstract:Objective: To explore the potential active compounds and possible target pathways of Wuwei Qingzhuo powder decoction in the treatment of functional diarrhea (FD) based on network pharmacology. Methods: Using TCMSP and ETCM database to obtain and screen the active ingredients of the addition and subtraction formula of Wuwei Qingzhuo Powder, query its structural information in PubChem, and use Swiss Target Prediction and Super Pred to predict the action Target of the drug. Drug Bank, Gene Cards and OMIM databases were used to retrieve FD targets. The intersection targets of drugs and diseases were selected to import STRING, and the target protein-target protein interaction network was drawn. Metascape was used for GO and KEGG enrichment analysis of key targets, and the action mechanism of addition and subtraction of Wuwei Qingzhuo powder and FD action target pathway were predicted. Results: A total of 371 FD related targets were collected, and 227 active component targets of Wuwei Qingzhuo powder addition and subtraction formula were involved in 51 FD targets. GO and KEGG enrichment analysis showed that wuwei Qingzhuo powder may act on SRC, PTPN11, PIK3R1, AKT1, MAPK1, STAT1, SYK and other targets, and achieve its goal of treating FD through immune-related pathways, inflammation-related pathways, smooth muscle expansion and other pathways. Conclusion: The network pharmacology proves that the addition and subtraction of Wuwei Qingzhuo powder can improve FD with the characteristics of multi-component, multi-target and multi-channel coordinated treatment of diseases, which provides theoretical support for the clinical application of The addition and subtraction of Wuwei Qingzhuo powder.
