摘    要:目的:本实验研究表没食子儿茶素没食子酸酯对阿米卡星诱导的大鼠听觉功能损害的保护作用。方法:设生理盐水对照组、Amk(450mg/kg.d×14d)损伤组和Amk加EGCG(10mg/kg.d,50mg/kg.d,100mg/kg.d×14d)保护组。五组动物各6只分别于试验前和试验的第7天、14天、21天以及35天行听觉脑干反应阈值检测。结果:第7天、14天、21天以及35天Amk损伤组跟对照组比较10kHz及20kHz阈移显著,10mg/kg EGCG保护组与损伤组比较10kHz及20kHz阈移不显著,损伤组与50mg/kg和100mg/kg EGCG保护组比较10kHz及20kHz阈移显著。结论:EGCG可以减轻阿米卡星诱导的ABR阈移,有效地抑制Amk所诱导的耳毒性。

(-)-Epigallocatechin-3-gallate protect Amikacin-induced hearing loss in rats
Abstract:Objective:To determine amikacin(Amk)-induced hearing loss in rats and to observe the protection of(-)-epigallocatechin-3-gallate(EGCG) in vivo.Methods: The study was comprised of one group serving as the saline control group,one group receiving Amk at 450mg/kg once a day for 14 days,three groups receiving Amk plus EGCG at 10,50,100mg/kg once a day for 14 days,respectively.The auditory thresholds were measured by evoked auditory brainstem responses.The thresholds were taken for each animal before and after amikacin application of 1 week,2 weeks,3 weeks and 5 weeks,respectively.Results: Saline-injected animals maintained stable thresholds throughout the course of test.In contrast,the animals receiving Amk developed a progressive hearing loss at both 10kHz and 20kHz.The auditory thresholds were remained elevated after cessation of treatment.The effect of EGCG on Amk-induced threshold shifts was dose dependent.No effect on Amk-induced threshold shifts was observed at 10mg/kg of EGCG once a day.A consistent attenuation at both 10kHz and 20kHz was observed with dose of 50mg or 100 mg/kg of EGCG once a day.Conclusion: Amikacin-induced auditory threshold shifts could be attenuated by EGCG.
