小鼠血浆中红花红色苷的RP-HPLC法分析及其药动学研究

摘    要:目的:建立小鼠血浆中红花红色苷的反相高效液相色谱(RP-HPLC)分析方法,并研究其药动学。方法:采用RP-HPLC法测定红花红色苷的血浆浓度,建立血药浓度-时间曲线,并得到主要的药动学参数。结果:红花红色苷的血浆浓度在0.558~55.8μg.L-1范围内与其峰面积积分值呈良好的线性关系(r=0.9992),最低检出限为0.005μg.L-1;红花红色苷在体内分布为二室模型,吸收快。结论:本方法简便、灵敏、重现性好,能满足药动学的研究需要。

Abstract:OBJECTIVE: To establish an RP- HPLC analysis of Carthamin in mice and to study its pharmacokinetics. METHODS: The serum concentration of Carthamin was determined by RP- HPLC.The blood concentration- time curve was established and the main pharmacokinetic parameters were computed .RESULTS: The linear range of Carthamin was 0.558- 55.8 μg · L^-1( r = 0.999 2), with the lowest limit of detection at 0.005μg · L^-1Carthamin in vivo assumed two- compartment model and rapid absorption. CONCLUSION: The proposed method is simple, sensitive and reproducible, and it met the standard for pharmacokinetic study.
