红花提取物对肾缺血再灌注损伤的影响

摘    要:目的:观察红花提取物对大鼠肾缺血再灌注损伤模型进行干预所产生的影响。方法:将40只大鼠随机分为4组(每组各10只),缺血再灌注组、假手术组、红花低剂量干预组和红花高剂量干预组。参照文献复制大鼠肾缺血再灌注损伤模型,并用不同浓度的红花提取物进行干预,之后取血检测血肌酐(Scr)、尿素氮(BUN)和丙二醛(MDA)的含量;取组织测定超氧化物歧化酶(SOD)和内皮素-1(ET-1)的含量;同时取肾组织进行HE染色观察病理学改变。结果:相对假手术组,缺血再灌注组、红花低剂量组和红花高剂量组大鼠的Scr、BUN、ET-1及MDA含量显著上升(P〈0.01),SOD显著下降(P〈0.01),肾单位出现明显变性坏死。红花高剂量组大鼠的Scr、BUN和ET-1值明显低于缺血再灌注组和红花低剂量组(P〈0.05);红花低剂量组和红花高剂量组大鼠的SOD值明显高于缺血再灌注组(P〈0.05),MDA值明显低于缺血再灌注组(P〈0.05)。经红花提取物干预的两组大鼠肾单位变性坏死较轻。结论:红花提取物可以减轻大鼠肾缺血再灌注损伤的程度、改善肾功能。

Abstract:Objective: To study the effect of safflower extraction on renal ischemia-reperfusion injury in rats. Methods: 40 rats were divided into 4 groups (n=10), including ischemia-reperfusion group, sham operation group, safflower low dose group and safflower high dose group. Renal ischemia-reperfusion injury model were established in rats, then treated with safflower in different doses. The blood was collected at the end of experiment to detect the expression of Scr, BUN, MDA, and renal tissues were collected to detect SOD and ET-1, and to observe the pathological changes in HE staining slides under mi- croscope. Results: Compared with sham operation group, Scr, BUN, ET-1 and MDA were increased significantly in the other three groups (P〈0.01), and SOD was decreased significantly (P〈0.01), meanwhile, serious degeneration and necrosis were observed in renal tissue. On the other hand, Scr, BUN, ET-1 in safflower high dose group were significantly lower than those in isehemia-reperfusion group and safflower low dose group (P〈0.05); furthermore, SOD in safflower low dose group and safflower high dose group were significantly higher than it in isehemia-reperfusion group (P〈O.05), but MDA was significantly lower than it in ischemia-reperfusion group (P〈O.05). The pathological changes in the two safflower treated groups were slighter. Conclusion: Safflower can alleviate the lesion of renal isehemia-reperfusion injury in rats, by which can improve the renal function.
