冻干血清培养基检测人染色体试剂盒的研究

摘    要:研制一种与传统培养液效果相当、不含小牛血清的外周血淋巴细胞培养液,并制成冻干试剂盒,用于染色体检测。通过批内和批间试验与传统培养液(对照组)比较,采用“转化淋巴细胞百分率”和“分裂指数”两项指标评估二者的应用效果,结果显示二者间差异均无显著性意义(P>0.05),淋转率变异系数实验组均小于对照组,提示试剂盒具有可靠性和稳定性。由于试剂盒成本低,培养时间仍为72小时,效果稳定,可完全替代传统培养液用于外周血染色体制备,尤适于基层推广应用。

Abstract:Abstract Objective :To study a culture solution of lymphocyte for peripheral blood without calf serum, and to make it a detection kit by freezing and drying. The effect of preparing the lymphocyte chromosome is same with the traditional culture solution. Melhods:Tocompare the kit with the traditional culture solution (campare group) by the test inside the same batch and between the different batchs. To employ two targets, "the percentage of the transforming lymphocyte" and "index of division", to estimate the effect of every group. Results: The shape and the size of the chromosomes in kit groups were good. There was no significant difference on the comparisons of two targets between the kit and traditional culture solution (P>0.05). and the coefficients of variation (CV) for "the percentage of the transforming lymphocyte"in kit groups were smaller than that of the control groups. Conclusion:The results pointed out that the effect of the kit without calf serum was reliable and stable. The steps of kit was simpler, the cost was lower, the time of culture was just 72h, It would be able to replace the traditional culture solution using in chromosome preparation for peripheral blood lymphocyte and it was especially suitable for spread and application in the grassroots units.
