福建省46株耐多药结核分枝杆菌katG基因突变特征研究

摘    要:目的:了解福建省耐多药结核分枝杆菌临床分离株katG基因的突变特点,为建立适合我省快速检测异烟肼耐药方法提供参考。方法:对福建省9个监测点进行耐药性调查,收集菌株经菌种鉴定、药物敏感性试验。选取46株耐多药结核分枝杆菌和15株全敏感株,扩增katG高突变区域的基因片段,测序后比对分析。结果:15株全敏感株未检测氨基酸突变。46株耐多药结核分枝杆菌katG基因的突变率为80.43%,katG 315的突变率为54.35%,联合突变率为52.17%。结论:福建省耐多药结核分枝杆菌杆不同耐药谱katG基因突变率不同,最常见的点突变发生于315位,463位具有等位基因多态性。

Abstract:Objective:To analyze the characteristic of katG gene mutation in the multi-drug resistant tuberculosis(MDR-TB) isolates from Fujian province,provide reference for building the suitable and fast method for detecting INH resistance in Fujian province.Methods: The strains were collected from nine drug resistant surveillance sites,and identification and drug susceptibility testing were performed.46 MDR-TB isolates and 15 pan-sensitive isolates were selected for producing the high mutation region in the katG gene by PCR,and analyzed by BLAST after DNA-sequencing.Results: No mutation was detected in 15 pan-sensitive isolates,the mutation rate of katG in MDR-TB isolates was 80.43%,the mutation rate of katG315 was 54.35%,and the united mutation rate was 52.17%.Conclusion: The mutation rate of katG was different in the different drug resistant spectrums of MDR-TB isolates.The most frequency mutation was katG315,and katG463 has polymorphism.
