拓扑替康对人卵巢癌顺铂耐药细胞株作用的研究

摘    要:目的:探讨拓扑替康(TPT)在体外诱导人卵巢癌顺铂耐药细胞株A2780/DDP凋亡的作用及其对凋亡相关蛋白表达的影响。方法:应用TPT诱导人卵巢癌顺铂耐药细胞株A2780/DDP凋亡,运用MTT法、透射电镜、DNA凝胶电泳检测和观察TPT对人卵巢癌顺铂耐药细胞株A2780/杀伤效应、凋亡形态、生物特性。采用Westernblot印迹杂交法以检测TPT对凋亡相关蛋白bcl-2和bax表达的影响。结果:TPT可诱导细胞株A2780/DDP凋亡,经TPT作用后A2780/DDP细胞内bax蛋白增加,而bcl-2蛋白不表达,bax/bcl-2比值增加。结论:TPT可诱导人卵巢癌顺铂耐药细胞株A2780/DDP凋亡,其机制可能与bax蛋白高表达及bax/bcl-2不表达有关。

Abstract:Objective:To boserve the effect of apoptiosis induced by topotecan(TPT) and study the experssion of apoptosis-related protein in cell lines A2780/DDP.Method:Apopotosis in cultured ovarian cancer cell lines A2780/DDP induced by TPT was investigated in vitro by applying MTT assay,electron microscope,DNA ladder so as to observe kill-efficience and apoptosis cell shape as well as biologhy characteristics.The method of western blot was used to test expression of apoptosis related protein bcl-2 and bax.Results:The cell lines A2780/DDP appeared typical characterisitic of cell apoptosis after being treat by TPT.The bax protein increased while bcl-2 protein expression was unclear.Conclusion:Our study showed that TPT were effective on inducing apoptosis of cisplatin-resistant human ovarian cancer cell lines A2780/DDP.One of the mechanism may be related to the high expression of bax protein and the increase bax/bcl-2 in ratio.
