雷公藤抑制免疫排斥反应的实验研究

摘    要:目的:通过雷公藤孵化皮片,在同种小鼠间进行移植,探讨雷公藤对免疫排斥反应的抑制作用。方法:采用15%雷公藤煎剂和40μg/ml、100μg/ml、500μg/ml雷公藤总甙对供鼠皮片浸泡孵化,在昆明种小鼠和DBA褐色小鼠间进行皮片移植,观察雷公藤对同种皮片移植免疫排斥的抑制作用。结果:雷公藤煎剂孵化皮片能显著延长移植物的存活时间,两种小鼠皮片移植后存活时间分别为(2231±360)天、(2150±340)天,与对照组(1320±225)天、(1181±275)天相比有显著意义(P<001);40μg/ml雷公藤总甙孵化后皮片存活时间为(1754±144)天,100μg/ml为(1984±263)天,500μg/ml为(2091±380)天;雷公藤总甙孵化加腹腔注射,每天40μg/20g小鼠为(2135±208)天,100μg为(2234±275)天,500μg为(1982±114)天,与对照组(1283±080)天、(1196±175)天相比均有显著差异(P<001);雷公藤总甙单纯孵化各组与孵化加腹腔注射相比,除40μg/ml组(P<005)外,存活时?

Abstract:Objective:To study the suppression of thunder godvine (tripterygiun wilfordii) to immunological rejective reaction during the transplant of isotype mice with hatched skin graft of thunder godvine. Methods:15% thunder godvine boiled solutions, and 40 μg/ml, 100 μg/ml, 500 μg/ml of thunder godvine glucoside were used to soak and hatch the mice skin graft, and then transplant the skin graft between Kunming-type mice and DBA brownish mice,so as to observe the suppression of thunder godvine to immunological rejection of isotype skin graft.Results: It is obvious that the hatched skin grafts of thunder godvine boiled solutions could prolong the surviving time of the hemotransplant, the surviving time of two kinds of mice was (22.31±3.60) days, (21.50±3.40) days respectively, there was significant difference ( P <0.01), as compared with the control group(13.20±2.25) days, (11.82±2.75) days. The skin graft survivaltime was (17.54±1.44) days.with 40 μg/ml thunder godvine glucoside eclosion; (19.84±2.63) days with 100 μg/ml eclosion; (20.91±3.80) days with 500 μg/ml eclosion. Every day 40 μg/20g of thunder godvine glucoside was used to hatch and inject the mice for (21.35±2.08) days; 100 μg for (22.34±2.75) days and 500 μg for (19.82±1.14) days.It had significant difference( P <0.01) as comparision of pure eclosion of each group of thunder godvine glucoside with the eclosion together with cavity injection except 40 μg/ml group ( P <0.05)], the survival period had no remarkable differences ( P <0.05). Conclusion:Thunder godvine colud obviously prolong the skin gragt survival period and obtain satisfactory results by was of simple eclosion。
