MTT法与流式细胞术检测肺炎合剂促进T淋巴细胞转化的实验研究

摘    要:目的:探讨肺炎合剂对小鼠T淋巴细胞转化的影响,并试图为药研究淋巴细胞转化提供检测方法依据。方法:通过病毒性肺炎患儿淋巴细胞转化率降低的机体,用氢化可的松建立免疫低下小鼠模型,以不同剂量药液灌胃给药后,采用MTTI法和流式细胞仪检测法对比观察肺炎合剂对ConA诱导的小鼠脾T淋巴细胞转化的作用。结果:肺炎合剂使正常和免疫低下小鼠脾T淋巴细胞增殖能力显著增强。结论:肺炎合剂对小鼠脾T淋巴细胞增反应的促进作用,可能是该药作用机理之一。并认为流式细胞术是测定淋巴细胞转化的一种准确而可靠的方法。

Abstract:Objective: In this article, the author inquires into the fact that pneumonia mixture has an influence over the transformation of T lymphocytes of small mice and tries to provide the basis of detective method for Chinese herbal medicine to study the transformation of lymphocytes. Methods: Through the organism of the lowering of transformation rate of lymphocytes of infantile patient with mimetic virus pneumonia, small mice model of hypoimmunity was setup with hydrocortisone, and with different dosages of liquid medicine, then poured it down into the stomach, MTF method and FCM apparatus were taken to detect and made a contrastive observation of the function of the transformation of spleen T lymphocytes of small mice induced by ConA. Results: Pneumonia mixture made the reproductive capacity of spleen T lymphocytes of the normal and hypoimmunity of the small mice obviously strengthen. Conclusion: That pneumonia mixture had an influence over the promoting function of productive reaction of spleen T lymphocytes of small mice might be one of the mechanism of the therapeutic function of this drug. It was considered that FCM technique was a kind of accurate and reliable method to determine the transformation of lymphocytes.
