NK细胞杀伤人胶质瘤起始细胞的体外研究

摘    要:背景与目的:越来越多的证据支持人脑胶质瘤来源于胶质瘤起始细胞/干细胞(glioma initiating cell/stemcell,GIC/GSC)的假说,鉴于干细胞特性.常规手术和放化疗难以清除GIC.本研究试图用体外活化的自然杀伤(natural killer,NK)细胞对GIC细胞进行杀伤研究。方法:以体外长期培养于干细胞培养基的CD133阳性胶质瘤细胞作为GIC,用免疫磁珠从胶质瘤患者(或健康人)的外周血中分离NK细胞,常规IL2/PHA活化NK细胞后,应用LDH释放法检测其体外杀伤GIC细胞的活性,以K562细胞为阳性对照。结果:体外实验中,异体NK细胞可以在GIC的培养基甩生长和活化.且其杀伤GIC细胞的能力随效靶比增高而增强,同一效靶比时,活化的NK细胞杀伤GIC细胞的活性明显高于未活化(静止)NK细胞的活性(P〈0.011。结论:异体NK细胞可以在GIC的培养环境中活化并表现出针对GIC的杀伤能力,为NK细胞用于脑胶质瘤起始细胞的临床清除提供可能性。
