| 摘 要： | 背景与目的：越来越多的证据支持人脑胶质瘤来源于胶质瘤起始细胞／干细胞（glioma initiating cell／stemcell，GIC／GSC）的假说，鉴于干细胞特性．常规手术和放化疗难以清除GIC．本研究试图用体外活化的自然杀伤（natural killer，NK）细胞对GIC细胞进行杀伤研究。方法：以体外长期培养于干细胞培养基的CD133阳性胶质瘤细胞作为GIC，用免疫磁珠从胶质瘤患者（或健康人）的外周血中分离NK细胞，常规IL2／PHA活化NK细胞后，应用LDH释放法检测其体外杀伤GIC细胞的活性，以K562细胞为阳性对照。结果：体外实验中，异体NK细胞可以在GIC的培养基甩生长和活化．且其杀伤GIC细胞的能力随效靶比增高而增强，同一效靶比时，活化的NK细胞杀伤GIC细胞的活性明显高于未活化（静止）NK细胞的活性（P〈0．011。结论：异体NK细胞可以在GIC的培养环境中活化并表现出针对GIC的杀伤能力，为NK细胞用于脑胶质瘤起始细胞的临床清除提供可能性。
