白头翁醇提物对葡聚糖硫酸钠诱导大鼠结肠炎肥大细胞致炎作用的抑制研究

摘    要:目的 研究白头翁醇提物对葡聚糖硫酸钠 (DSS)诱导结肠炎肠粘膜肥大细胞 (IMMC)致炎作用的抑制效果。方法 建立 DSS诱导的大鼠结肠炎模型 :SD大鼠 2 8只 ,随机分为三组 :白头翁治疗组 (1 0只 ) 1 - 7天用白头翁醇提物2 g/ kg/ d用蒸馏水配制成 1 ml溶液每天灌肠一次 ,自由饮用 3%DSS溶液 ;模型组 (1 0只 ) 1 - 7天用等同的蒸馏水 1 ml每天灌肠一次 ,自由饮用 3%DSS溶液 ;对照组 (8只 ) 1 - 7天用等同的蒸溜水 1 ml每天灌肠 1次 ,自由饮水。观察大鼠的腹泻、便血症状及结肠组织学改变。分别用莹光法测定结肠组织组胺浓度 ,EL ISA测定结肠组织肿瘤坏死因子 -α(TNF-α)和前列腺素 E2 (PGE2 )水平。组织化学法 IMMC染色并记 IMMC数。结果 白头翁治疗组与模型组比较 ,治疗组大鼠的症状减轻结肠粘膜组织损伤显著改善 (P<0 .0 5 ) ,结肠组织组胺水平升高 ,TNFa和 PGE2显著降低 (P<0 .0 1 )。 IMMC计数减少 (P<0 .0 5 )脱颗粒现象减弱。结论 白头翁醇提物具有明显的抗炎作用 ,其机制是通过抑制 IMMC的活化和组胺的释放 ,终止肥大细胞 -细胞因子级联反应 ,降低 TNFa和 PGE2生成 ,从而减轻结肠炎性损伤和腹泻、便血等症状。

Inhibiting effect of radix pulsatillae alcohol extract on the activity of intestinal mucosal mast cells in dextran sulfate sodium induced colitis of rats

Abstract:Objective To investigate the suppressive effect of radix pulsatillae alcohol extract on the activity of intestinal mucosal mast cells (IMMC) in dextran sulfate sodium (DSS) induced colitis of rats. Methods Acute colitis was induced by giving 3% DSS orally. 28 male rats were randomized into radix pulsatillae alcohol extract-treated group(group A, n=10), DSS control group(group B, n=10) and normal control group(group C, n=8). The rats in group A were perfused through colon with radix pulsatillae alcohol extract at the dosage of 2g/kg/d for 1 ml solution from day 1 to day 7 and drank 3% DSS solution. The rats in group B were treated with distilled water in an equal volume as group A and drank 3% DSS solution from day 1 to day 7. The rats in group C were treated with distilled water as group B from day 1 to day 7 and drank water normally. Diarrhea and bloody stool as well as colonic histology were observed. The levels of colonic tissue histamine, tumor necrosis factor a(TNFa) and prostaglandin E2(PGE2) were determined by fluorimetry and ELISA, and the expression of IMMC in colonic tissue was detected by histochemical stain method. Results Compared with DSS control group, the inflammatory symptoms and histological damages of colonic mucosa in radix pulsatillae alcohol extract-treated group were significantly improved (P<0.05). The level of histamine in colonic tissue was significantly elevated (P<0.05). The tissue levels of TNFa, PGE2 and the expression of IMMC in colonic tissue were significantly reduced (P<0.05-P<0.01). Conclusion The data suggest that radix palstillae alcohol extract has an anti-inflammatory action on experimental colitis by reducing the tissue levels of TNFa, PGE2 and suppressing IMMC activity in colonic tissues.
