酸味中药复方对2型糖尿病大鼠ET-1、NO及肾脏ICAM-1基因表达的影响

摘    要:目的:观察酸味中药复方对2型糖尿病大鼠血浆内皮素-1(ET-1)、血清一氧化氮(NO)及肾脏细胞间黏附因子-1(ICAM-1)基因表达的影响,探讨该方保护糖尿病大鼠肾脏病变的作用机制。方法:Sprague-Dawley雄性大鼠采用链脲佐菌素(STZ)配合高热量饲料复制2型糖尿病模型,将成模大鼠分为中药组(SFCC组)、氨基胍组(AG组)、模型组(T2DM组)及正常组(NC组),用药干预8周和12周后,采用放免法和硝酸还原酶法检测血浆ET-1和血清NO含量,原位杂交法检测大鼠肾脏ICAM-1的基因表达。结果:T2DM组ET-1含量及肾脏ICAM-1基因表达明显高于NC组(P〈0.05),NO明显低于NC组(P〈0.05);SFCC组和AG组ET-1含量及肾脏ICAM-1基因表达明显低于T2DM组(P〈0.05),NO明显高于T2DM组(P〈0.05)。结论:酸味中药复方可能通过调节ET-NO,抑制ICAM-1基因表达实现其对糖尿病大鼠肾脏病变的保护作用。

Abstract:Objective:To investigate the possible mechanism of Sour-Flavor Chinese Compound (SFCC) in protecting renal lesions of type 2 diabetic mellitus (T2DM) rats, by analyzing the effects of SFCC on plasma endothelin-1 (ET- 1 ) ,serum nitric oxide(NO) and the expression of ICAM- 1 in nephridial tissues in T2DM rats. Methods:T2DM rats, which were fed with highcaloric diets, were injected with streptozotocin(STZ). The rats were randomly divided into SFCC group, AG group and T2DM group. At the 8th weeks and 12th weeks after medication, the ET - 1 was determined by radio - immunity, the serum NO was determinedby nitrate reductase,and the expression of ICAM-1 mRNA in nephridial tissues was detected by situ hybridization. Results: The ET- 1 and positive expression of ICAM - 1 mRNA in nephridial tissues of T2DM group were higher than the normal group (P〈0.05), and the serum NO was lower than normal group( P〈0.05). The ET-1 and positive expression of ICAM- 1 mRNA in nephridial tissues in SFCC group and AG group were lower than T2DM group (P〈0.05),and the serum NO were higher than T2DM group( P 〈 0.05 ). Conclusion: Sour - flavor Chinese compound inhibits the positive expression of ICAM - 1 mRNA through the regulation of ET - NO. That may be the mechanism of its protection of T2DM rats renal lesions.
