固体脂质纳米粒降低雷公藤内酯醇肝毒性的实验研究

摘    要:目的 探讨固体脂质纳米粒降低雷公藤内酯醇(TP)对肝脏的毒性作用。方法 用雷公藤内酯醇固体脂质纳米粒(TP—SLN)小鼠ig60d后,测定小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和肝组织中的超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH—Px)的活性以及丙二醛(MDA)的含量。结果与空白组相比,TP—SLN对ALT,AST,SOD,GSH—Px,MDA的活性或含量差异无显著性;中、高剂量(20,30μg/kg)的TP组则显著降低SOD和GSH—Px的活性,并能显著提高MDA,ALT和AST的含量。结论 TP—SLN能降低TP对肝脏的毒性。

Abstract:Object To study the mechanism of triptolide solid lipid nanoparticle(TP-SLN) for decreasing liver toxicity of triptolide(TP).Methods With ig three doses of TP-SLN to mice for 60 d,the ALT,AST activities in serum and superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) activ ites and malondialdehyde(MDA) contents in liver were determined.Results The activities of ALT,AST,SOD,GSH-Px and content of MDA between experimental group and blank group did not have remarkable difference.However,the activites of ALT,AST,SOD,GSH-Px for the middle-(20 g/kg) and high-dose(30 g/kg) group were higher and the contents of MDA were lower than t he experimental group.Conclusion TP-SLN can decrease the liver toxicity of TP.
