视网膜缺血再灌注后c-fos、c-jun蛋白表达

摘    要:目的:观察大鼠视网膜缺血再灌注(RIR)后,凋亡即刻基因cfos、cjun在视网膜各层细胞中的表达变化,进一步明确凋亡即刻基因和RIR损伤发生机制的关系。方法:采用视网膜中央动脉结扎方法诱导大鼠视网膜缺血60min,解除结扎,建立RIR模型。缺血60min,再灌注0,1,3,6,12,24h作视网膜石蜡切片,cfos,cjun免疫组化观察。结果:正常对照组和缺血组未见cfos、cjun表达,RIR后,视网膜神经节细层(ganglioncelllayer,GCL)和内核层(innernuclearlayer,INL)1h开始少量表达(P<0.05),3h达到高峰(P<0.05),6h开始下降(P<0.05),24h几乎趋于正常组表达(P>0.05)。而外核层(outernuclearlayer,ONL)几乎无阳性表达。结论:cfos、cjun参与RIR损伤发生,RIR中视网膜节细胞层和内核层细胞存在凋亡征象,尤以3h组最为显著。

Abstract:Objective: To determine whether there is a change in c-fos and c-jun immediate early gene expression in retina after retinal ischemia reperfusion in rats. Methods: Retinal ischemia was induced in SD rats by means of ligation of the retinal central artery for 60 minutes. The model of retina ischemia reperfusion was established once releasing the ligation. The expression of c-fos and c-jun in the paraffin section of retina was observed at the beginning(0 hour), and the 1st, 3rd, 6th, 12th, 24th hours after 60-minute-ischemia by immunohistochemical method. Results: The expression of c-fos and c-jun first appeared in the ganglion cell layer and inner nuclear layer at the 1st hour (P<0.05), but not in the outer nuclear layer. At the 3rd hours, the expression of c-fos and c-jun reached to the peak(P<0.05), and then decreased gradually at the 6th hour (P< 0.05). At the 24th hours, the expression of c-fos and c-jun arrived to the normal level (P> 0.05). Weak signals for c-fos and c-jun were observed in the normal group and the 0-hour group. Conclusion: It suggests that changes exist in expression of c-fos and c-jun after retinal ischemia reperfusion, and the apoptosis is obvious in the ganglion cell layer and inner nuclear layer after the retinal ischemia reperfusion injury, especially in the group of retina ischemia reperfusion after three hours.
