摘    要:目的:研究p16 mRNA在鼻咽癌组织中的表达情况,探讨p16 mRNA与鼻咽癌生物学行为的关系。方法:应用生物素标记RNA探计的原位杂交技术,对20例散发性鼻咽癌(NPC)患者及3个NPC家系中NPC患者的组织标本进行mRNA检测,探讨p16 mRNA的异常表达在NPC的发生发展中的作用,以20例鼻咽炎患者的鼻咽部组织做为对照。结果:20例NPC标本原位杂交阳性8例,p16 mRNA阳性表达为40%(8/20),3个高发家系的NPC标本无阳性表达,20例对照(鼻咽炎患者的标本)全部有p16 mRNA阳性表达。结论:p16 mRNA在鼻咽癌组织中呈低表达,提示p16基因作为一种重要的抑癌基因,它的突变或缺失可能与鼻咽癌的发生发展及恶性行为起一定的作用。

Abstract:Objective To investigate the expression of p16 mRNA in the tissue of nasopharyngeal carcinoma(NPC) patients and the relationship between p16 mRNA and the biological behavior of NPC. Methods The mRNA of the tissue samples of the 20 sporadically distributed NPC cases and the NPC patients of NPC families were examined by in situ hybridization by using RNA probe labeled by with biotin, another 20 inflammatory nasopharyngeal mucosa patients were taken as control. Results Out of the 20 NPC patients 8 were positive in in situ hybrization with a positive rate pf expression of mRNA was 40%(8/20). No positive results were observed in the 3 NPC patients of NPC families. While all the cases in the control group were positive. Conclusion The results of this study illustrate that the mRNA was is lowly expressed, suggesting that the mutation and deletion of p16 which is taken as a suppressing gene may play a certain role in the genesis and development of NPC and biological behavior.
