弓形虫速殖子和包囊的分离纯化

摘    要:采用植物血凝素P(PHAP)亲和凝集分离法自小鼠腹腔液内分离弓形虫速殖子,纯度达99.5%,收获率为73.2%。与胰蛋白酶消化法相比,前法对虫体无损伤。此外,应用淋巴细胞分离介质Ficoll-Paque密度梯度离心法自鼠脑组织内分离出较纯净的包囊。

Abstract:Toxoplasma gondii tachyzoites were separated from mouse peritoncal exudates by using a lectin of phytohemagglutinin P. A purity of 99.5% and a recovery of 73.2% were obtained and no chemicaldamage to the parasite surface protein was found when compared with trypsin digestion procedure by SDS-polyacrylamide gel clectro-phoresis. Besides, cysts were isolated from mouse brain tissues with density-gradient centrifugation on Ficoll-Paque, a lymphocyte isolation liquid.
