去铁酮大鼠血浆蛋白结合率的实验研究

目的建立测定透析液中去铁酮（DFP）浓度的高效液相色谱法（HPLC），观察DFP的大鼠血浆蛋白结合率。方法以体外方式，采用平衡透析法模拟DFP在大鼠体内与血浆蛋白结合的过程，并用HPLC法测定其透析液中DFP的浓度及血浆蛋白结合率。结果低、中、高3种浓度下，DFP的大鼠血浆蛋白结合率范围为4.25%～5.31%，不同浓度组血浆蛋白结合率差异无统计学意义（P＞0.05）。结论DFP血浆蛋白结合率较低，属于低血浆蛋白结合药物，其主要以游离形式存在而发挥药效。

Plasma protein binding rate of deferiprone in rats

Objective To develop a HPLC method for determination of deferiprone(DFP) concentration in dialysate and to study the plasma protein binding rate of DFP. Methods With an external manner, we used an equilibrium dialysis method to imitate the binding process between DFP and plasma protein in vivo and determine the concentration of DFP and the plasma protein binding rate of DFP. Results The plasma protein binding rate of DFP at low, middle and high concentrations was between 4.25% to 5.31% , and it was not significantly different in all groups (P > 0.05). Concolusion The plasma protein binding rate of DFP is low. So DFP is a kind of low plasma protein binding rate drug, and most of the drug molecules act on a body with a free type.
