| 摘 要： | 目的:探讨核糖核苷酸还原酶M2(RRM2)在人胆管癌组织及胆管癌细胞系中的表达、分布以及生物学意义。方法:采用免疫组织化学技术(SP法)检测RRM2基因和蛋白在40例胆管癌标本、40例癌旁胆管组织以及10例正常胆管组织标本中的表达;同时采用RT-PCR法及Western blot法分别检测了RRM2基因mRNA及其蛋白在人胆管癌细胞系QBC939以及人正常胆管上皮细胞系HIBEC中的表达。结果:RRM2基因蛋白在胆管癌组织中表达阳性率为72.5%,而在癌旁胆管组织和正常胆管组织中未能检测到RRM2表达。在人胆管癌细胞系QBC939中,RRM2的mRNA及蛋白均特异性表达,而在正常人胆管上皮细胞系HIBEC中未见RRM2的表达。结论:RRM2在人类胆管癌组织和胆管癌细胞系中选择性高表达,可能与胆管癌的发生、发展密切相关。
