过敏性紫癜患儿治疗后早期外周血T淋巴细胞上PD-1的表达

摘    要:目的探讨过敏性紫癜患儿经抗过敏治疗后早期外周血T淋巴细胞上PD-1分子的表达变化及其与体液免疫IgA的关系。方法应用荧光抗体染色技术经流式细胞仪动态分别检测20例健康对照组和40例过敏性紫癜患儿急性期和经抗过敏治疗后早期外周血T淋巴表面的CD4+和CD8+T细胞表面的PD-1分子的表达水平,同时应用免疫比浊法查血清IgA水平,探讨其表达与血清IgA水平之间的关系。结果过敏性紫癜患儿治疗前急性期和与健康对照组相比,外周血T淋巴表面的CD4+T细胞组PD-1在外周血淋巴细胞阳性表达率明显升高,差异有统计学意义(P<0.01);外周血T淋巴表面的CD8+T细胞组PD-1在外周血淋巴细胞阳性表达率升高,差异有统计学意义(P<0.05);同时,血清IgA水平升高,差异有统计学意义(P<0.05);治疗后与健康对照组相比,差异无统计学意义(P>0.05)。结论有效的抗过敏治疗可使过敏性紫癜患儿急性期外周血清IgA水平逐渐降低,T淋巴细胞表面PD-1表达下降,推测IgA水平与T淋巴细胞表面表面PD-1表达水平变化与患儿免疫调节机制紊乱有关。

Abstract:Objective To investigate the expressions of PD-1 on peripheral lymphocytes after anti-allergy early treatment from patients with Henoch-Schonlein purpura(HSP)and its relationship with serum IgA.Methods Peripheral lymphocytes and IgA were obtained from 40 patients with HSP(HSP group)and 20 healthy human controls(control group).Immunofluorescent staining and flow cytometry were used to measure the expressions of CD4 + and CD8 + T cell PD-1 on peripheral lymphocytes.Immunoturbidimetric method was used to detect the serum levels of IgA to explore the relationship between IgA and the expressions of CD4 + and CD8 + T cell PD-1 on peripheral lymphocytes.Results The expression rate of CD4 + and CD8 + T cell of PD-1 on peripheral lymphocytes significantly increased in HSP group of acute phase compared with those in the control group(P<0.01).At the same time,serum IgA levels were increased,the difference was statistically significant(P<0.05);No significant difference was revealed in the expression of IgA between HSP group after treatment and control group(P>0.05).Conclusion The effective anti-allergy treatment of children with Henoch-Schonlein purpura can reduce the acute peripheral serum IgA levels and PD-1 on the T-lymphocyte surface expression.It is suggesting that IgA levels and T cell surface expression of PD-1 surface changes would be relationed with children immune system disorders.
