摘    要:目的优化重组人白细胞介素-18的发酵工艺,以获得工程菌的高密度发酵和目的蛋白的高表达.方法在10L发酵罐中,研究重组人白细胞介素-18工程菌在不同的pH值、溶氧和诱导时间对工程菌生长和目的蛋白表达的影响.结果根据优化条件,连续三批重复发酵10L工程菌,最终菌体密度均达A600nm=28,菌体获得量均可达湿重50g?L-1以上.目的蛋白表达量均为40%以上.结论优化了重组人白细胞介素-18基因工程菌的发酵和表达条件,为规模化生产奠定了基础.

Abstract:OBJECTIVE To optimize the fermentation procedure of recombinant human interleukin-18 and to obtain high cell density and high level expression of the aim protein.METHODS Study the influence of the rang of pH,dissolved oxygen and induction time on the growth of Engineered E.coli and expression of aim protein in a 10L fermentor.RESULTS Under the established conditions,we performed a three-time repeated fermentation .The final cell density was A600nm=28 and more than 50g of wet bacteria per liter could be obtained.The expression level of aim protein was higher than 40%.CONCLUSION The condition for fermentation and expression of recombinant human interleukin-18 was optimized.It laid a foundation of large-scale production of recombinant human interleukin-18.
