摘    要:目的:探讨fibulin糖蛋白家族中fibulin-3(EFEMP1)对卵巢癌细胞增殖、侵袭与转移的影响,为卵巢癌针对性治疗提供理论基础。方法:利用慢病毒转染RNA干扰的方式,转染具有高侵袭转移能力的S1克隆细胞株,抑制EFEMP1表达。Western blot、Real-time RT-PCR和免疫组化对转染效果进行验证。采用体外细胞功能分析技术检测干扰前后EFEMP1对细胞增殖、侵袭与转移能力的影响。建立裸鼠皮下移植瘤模型,体内观察EFEMP1对肿瘤生长的影响。结果:RNA干扰有效抑制EFEMP1表达,EFEMP1降表达显著抑制肿瘤细胞增殖、侵袭和转移能力,裸鼠体内成瘤率降低并且肿瘤体积显著下降,同时肿瘤组织Fibulin基因表达显著降低。结论:EFEMP1与卵巢癌发生发展及侵袭转移密切相关,为卵巢癌的治疗提供新的思路。

Effect of EFEMP1 on invasion,metastasis and proliferation of human ovarian cancer cell
Abstract:Objective:To explore the effect of (Fibulin-3) EFEMP1 on invasion,me-tastasis and proliferation of human ovarian cancer cell, which belongs to fibulin glycoprotein family,the related results will provide theoretical basis for ovarian cancer targeted therapy. Methods:The highly invasive cell line S1 was infected with lentivirus vector with EFEMP1 shRNA. Real-time RT-PCR,Western blot and ICC were performed to authenticate transfection effect . The vitro cell function analyses were used to study the effect of EFEMP1 on cell prolifer-ation,invasion and metastasis. Furthermore, tumor xenografts in nude mice were performed to observe the role of EFEMP1 in the progress of ovarian cancer in vivo. Results:Knockdown of EFEMP1 significantly inhibited EFEMP1 expression,down-regulated EFEMP1 suppressed tumor cell proliferation, invasion and metastasis abilities, and decreased the tumor forming rate and volume in nude mice. Conclusion:EFEMP1 is related to ovarian cancer development and pro-gression,and may serve as a new therapeutic target for the treatment of ovarian cancer in the fu-ture.
