摘    要:目的建立HPLC法测定葛花中大豆苷、染料木苷的含量。方法采用C18色谱柱(250mm×4.6mm,5μm);流动相为乙腈-1.0mL·L-1磷酸溶液(15∶85);流速为1.0mL·min~(-1);检测波长为260nm;柱温为30℃。结果大豆苷回归方程为Y=166 989.8 X-1 438.6,r=0.999 9,大豆苷在0.206~1.032μg范围内线性关系良好;染料木苷回归方程为Y=142 567 X+2 543.1,r=0.999 7,染料木苷在0.204~1.018μg范围内线性关系良好。结论该方法简便、准确,重复性好,可作为葛花的质量评价依据。

Determination of daidzin and genistin glycosides in the flower of kudzuvine by HPLC
Abstract:Objective To establish an HPLC method for the determination of the content for daidzin,genistin glycosides in the flower of kudzuvine.Methods C1 8 column (250 mm×4.6 mm,5 μm)was used with acetonitrile-1 mL·L-1 phosphoric acid (1 5∶85)as the mobile phase;the flow rate was 1.0 mL·min-1 ,the detection wavelength was 260 nm,and the column temperature was 30 ℃. Results Daidzin regression equation was Y = 1 66 989.8X - 1 438.6,r =0.999 9,indicated that daidzin injection amount between 0.206-1.032 μg had good linear relationship;genistein glycosides regression equation was Y = 142 567X +2 543.1,r =0.999 7, showed that genistein glycosides injection amount between 0.204-1.018 μg had good linear relationship.Conclusion The method is simple,accurate,reproducible,and can be used as a quality assessment method for the flower of kudzuvine.
