毁损伏隔核对吗啡给药大鼠条件性位置偏爱与脑功能的影响

摘    要:目的:探讨双侧毁损伏隔核对吗啡给药大鼠条件性位置及大鼠脑功能的影响。方法:6-羟多巴胺注射毁损大鼠双侧伏隔棱,以条件性位置偏爱实验评价伏隔核对大鼠的影响;以Morris水迷宫实验评价对空间学习记忆功能的影响。结果:在条件性位置偏爱实验中,单纯给药组与毁损给药蛆、单纯毁损组、对照组相比较存在显著差异(P〈0.05)。在自然戒断状态下。单纯给药组条件性位置偏爱先增加后减少。水迷宫实验毁损组与对照组相比,毁损前后自身对照均无明显差异(P〉0.05)。结论:伏隔核中的多巴胺系统在精神依赖(大鼠表现为条件性住置偏爱)形成过程中起关键性作用。

Abstract:Objective: To explore the effect of bilateral nucleus accumbens(NA) lesion on conditioned place preference(CPP) in rats receiving morphine and on the brain function of rats.Methods: 6-hydroxydopamine was slowly injected into bilateral NA of rats with NA lesion,and saline was injected into the rats without NA lesion.The CPP and spatial memory of the rats were determined by using CPP test and Morris water maze test,respectively.Results: In CPP test,there were significant differences in CPP between the morphine alone group and other 3 groups,impaired alone group,impaired plus morphine group,and control group(P<0.05).CPP increased obviously at first and then decreased during the process of natural withdrawal in morphine alone group,In Morris water maze test,there was no significant difference between the impaired group and control group and between the rats before and after impairment in impaired group(P>0.05).Conclusion: Chemically impaired dopamine systems in NA of rats play an important role in the formation of psychological dependence.
