槲皮素对人骨肉瘤细胞U-2OS/MTX300增殖和凋亡的影响及其机制研究

摘    要:目的:研究槲皮素(quercetin,Qu)对骨肉瘤细胞系U-2OS/MTX300增殖和凋亡的作用及其机制。方法:MTT法观察细胞增殖活性;Annexin V/PI染色检测凋亡;线粒体膜电位及细胞色素C的Western blot检测线粒体凋亡途径;持续活化Akt瞬时转染、Western blot检测Akt通路相关蛋白表达水平变化。结果:槲皮素可明显抑制耐甲氨蝶呤骨肉瘤细胞U-2OS/MTX300生长,并呈时间和剂量依赖性;Annexin V/PI可明显检测到细胞凋亡;进一步发现其作用机制是通过下调线粒体膜电位,促进细胞色素C向胞浆释放及抑制Akt磷酸化来实现。结论:槲皮素可显著抑制耐甲氨蝶呤骨肉瘤细胞系U-2OS/MTX300细胞增殖并诱导其凋亡,其机制与线粒体凋亡途径及抑制Akt活性有关。

Abstract:Objective: To study the effect and mechanisms of quercetin(Qu) on proliferation and apoptosis of human methotrexate resistant osteosarcoma cell U-2OS/MTX300. Method: MTT assay was used to observe cell proliferation. The apoptosis was examined by using Annexin V/PI staining. Western blot of mitochondrial membrane potential and cytochrome c were used to detect mitochondria spoptosis pathway. The protein expressions related to Akt pathway was detected by continuous activated Akt transient transfection and western blot. Result: Qu can obviously inhibit the growth of human MTX resistant osteosarcoma cell U-2OS/MTX300 cells in a dose- and time-dependent manner.Annexin V/PI staining showed obvious cell apoptosis. Reduction of mitochondrial membrane potential, release of mitochondrial cytochrome c to cytosol and dephoshorylation of Akt were observed after Qu treatment. Conclusion: Qu can inhibit proliferation and induce apoptosis of human MTX resistant osteosarcoma cell U-2OS/MTX300, which may be related with mitochondrial apoptosis pathway and Akt activity.
