环磷酰胺诱导建立少精/无精症大鼠模型所致睾丸、附睾IGF—Ⅰ和bFGF的变化

摘    要:目的 探讨环磷酰胺(CP)对大鼠附睾的组织中胰岛素样生长因子Ⅰ(IGF-Ⅰ)和碱性成纤维细胞因子(bFGF)表达的影响。方法 96只8周龄成年雄性SD大鼠,分为6组,每组16只。第1~5组分别给予CP10、20、40、80、100mg/kg·d,第6组为对照组,给予生理盐水2ml。胃管给药2周和4周后,处死大鼠,采用ELISA法对附睾组织洗脱液IGF-Ⅰ和bFGF进行定量检测;同时采用SP免疫组化法检测睾丸组织IGF-Ⅰ和bFGF的表达。结果 随着CP处理时间的延长和剂量的增加,IGF-Ⅰ和bFGF在大鼠附睾中的浓度逐渐下降,大鼠睾组织睾中IGF-Ⅰ和bFGF的表达也降低。结论 CP在引起大鼠少精子/无精子症的同时,可以显著降低大鼠附睾、睾丸IGF-Ⅰ和bFGF的含量和表达水平。IGF-Ⅰ和bFGF的降低与CP降低大鼠生精功能明显相关。

Abstract:Objective To evaluate the effect of cyclophosphamide on the expression of insulin-like growth factor-I (IGF- I) and basic fibroblast growth factor (bFGF) in the testes and epididymides in the rats. Methods Ninety-six male adult rats (8 weeks old) were divided into 6 groups of 16 rats each. The cyclophosphamide of 10, 20, 40, 80 and 100 mg/ (kg'd) was given to the rats respectively in groups 1, 2, 3, 4 and 5. The group 6 serv as control received 2ml 0. 9% NaCl. All the rats were sacrificed and the levels IGF-I and bFGF in the epididymides and their expressions in the testes were determined by ELISA techniques and immunohistochemieal SP technique respectively 2 and 4 weeks after the administration of the drug. Results The levels of IGF-I and bFGF in the epididymides and their expression in the testes were gradually reduced with the increase in time after administration of the drug and dose of the drug in the rats. Conclusions Cyclophosphamide not only causes oligozoospermia or azoospermia, but also produces a significant decrease in the levels of IGF-I and bFGF of the epididymides and the expression of IGF-I and bFGF of the testes in the rats. The decrease in IGF-I and bFGF of testes and epididymides may be relative to the low spermatogeneous function of the testes caused by the administration of cyclophosphamide.
