羟基磷灰石陶瓷结晶度对成骨细胞活性的影响研究

摘    要:目的:通过检测生长在羟基磷灰石(HA)陶瓷表面成骨细胞的碱性磷酸酶,来评价不同结晶度HA陶瓷对成骨细胞活性的影响,为该材料的临床应用提供实验依据。方法:在不同温度条件下烧结羟基磷灰石制成不同结晶度的HA陶瓷。通过免疫细胞化学方检测生长在不同结晶度HA表面成骨细胞碱性磷酸酶的表达。结果:在早期,结晶度高的HA陶瓷(烧结温度在1200℃和1000℃)比结晶度低的HA(烧结温度在800℃)诱导表达更多的碱性磷酸酶蛋白。然而,生长在结晶度高的HA陶瓷上的成骨细胞分泌碱性磷酸酶蛋白的量随着培养时间延长逐渐降低,一段时间后接近生长在结晶度低的HA陶瓷上的成骨细胞分泌碱性磷酸酶蛋白的量。结论:HA陶瓷结晶度通过调节成骨细胞碱性磷酸酶蛋白的表达调控成骨细胞的活性。

Abstract:Objective:To evaluate the effect of different crystallinities of hydroxyapatite(HA) ceramics on osteoblast activity via detecting alkaline phosphatase(ALP) protein in osteoblasts grown on HA ceramic surface.Methods:HA ceramics with different crystallinities were fabricated by sintering HA at different temperatures.ALP in osteoblasts grown on the HA ceramic surface was detected with immunohistochemistry.Results:In the early stage,HA ceramics with high crystallinities(sintered at 1200℃ and 1000℃) induced more ALP protein production than that of low crystallinity(sintered at 800℃).However,the ALP production decreased gradually with time in osteoblasts grown on the high crystallized HA ceramics and finally after 12 days,reached the level similar to that in osteoblasts on low crystallized HA ceramics.Conclusion:HA ceramic crystallinity can regulate the bioactivity of osteoblasts by controlling ALP protein production.
