摘 要: | 目的研究白细胞介素-18(interleukin 18,IL-18)基因单核苷酸多态性与河北唐山地区人群2型糖尿病(T2DM)的相关性。方法应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对122名T2DM患者和161名健康对照者(NC)行IL-18启动子-607C/A基因型检测,并测定血清hs-CRP。结果糖尿病与对照组比较,IL-18基因-607C/A多态性分布差异有统计学意义(P〈0.05),C等位基因携带者患T2DM的风险是A等位基因的1.883倍(OR=1.883,95%CI:1.28-2.771)。两组人群中CC和CA基因型个体CRP浓度均显著高于AA基因型个体(P〈0.05)。结论IL-18基因-607C/A多态性与T2DM的发病具有相关性,其中C等位基因可能是T2DM的遗传易感基因,并导致血清CRP浓度升高。

Abstract: | Objective To investigate the association of-607 C/A polymorphism in IL-18 promoter gene with type 2 diabetes mellitus(T2DM) in Tangshan population.Methods IL-18-607C/A polymorphism was detected by polymerase chain reaction-restriction fragment-length polymorphism in 122 type 2 diabetic subjects and 161 health subjects.The serum hs-CRP levels of patients and control subjects were measured by routine method.Results The IL-18 gene-607C/A polymorphism was significantly defferent between the T2DM and control group(P〈0.05).The realtive risk of T2DM for-607C allele carriers was 1.883 times of the-607A allele carriers(OR=1.883,95%CI: 1.28-2.771).Both patienst and control subjects with CC and CA genotypes have significantly higher IL-18 levels than those with AA genotype(P〈0.05).Conclusion The IL-18 gene-607 C/A polymorphism was associated with T2DM,the-607C allele may be a risk factor for T2DM.Moreover,the-607C allele may result in the enhanced production of IL-18 protein in patienst and control subjects. |
