摘    要:目的探讨组蛋白去乙酰化酶(HDAC)抑制剂——磺胺酰基苯胺复合物(SAC)对人膀胱癌细胞的增殖及凋亡作用及机制。方法以不同浓度的SAC作用于体外培养的膀胱癌细胞株UMUC3;采用MTr法检测细胞生长抑制率;以流式细胞术测定膀胱癌细胞的凋亡及细胞周期;应用RT-PCR检测膀胱癌细胞中的P21^waf/cifl mRNA。结果SAC有效抑制膀胱肿瘤细胞增殖,显著诱导细胞发生调亡且主要诱导G1期细胞发生调亡,显著诱导上调P21^waf/cifl mRNA表达。上述结果都呈现出显著的量-效与时-效关系。结论SAC在体外能有效抑制人膀胱癌细胞生长,可能是通过上调P21^waf/cifl mRNA水平实现的。

Abstract:Objective This study was designed to investigate whether histone deacetylase inhibitors ( sulfonamide anilides compound temporarily referred compound, SAC) possesses antitumor activity HumAn bladder cancer cell and if any, its mechanism. Methods Human bladder cancer UMUC3 cell lines was treated with different concentrations of SAC for different periods of time. After treatment, cell growth was measured by MTT assay. Cell apoptosis and changes in cell cycle were examined by means of flow cytometry (FCM). P21^waf/cifl mRNA expression was assessed by RT-PCR. Results SAC treatment caused cell arrest at G1 phase as shown by FCM and induced P21^waf/cifl mRNA expression which results were clearly displayed in the volume - with effect - effect relationship. Conclusion SAC is able to inhibit human bladder cancer cell lines growth in vitro, most likely through induction of P21^waf/cifl mRNA expression and subsequent arrest of cell cycling at G1 phase.
