雪旺细胞源神经营养因子对脊神经根性撕脱所致运动神经元死亡的保护作用

摘    要:目的研究雪旺细胞源神经营养因子(SDNF)对臂丛神经根性撕脱所致前角运动神经元死亡的保护作用。方法于颈神经根性撕脱处应用SDNF,3周后观察损伤侧脊髓前角运动神经元的存活率和形态学变化以及NOS表达的情况。结果对照组68.6%的神经元死亡,存活神经元胞体明显萎缩,同时表达NOS神经元增多;实验组的死亡率较对照组降低35%,存活神经元胞体无萎缩,表达NOS神经元未见增多。结论SDNF对脊髓前角运动神经元死亡有明显的保护作用,NO在臂丛神经根性撕脱所致运动神经元死亡中起一定作用。

Abstract:Objective To study the protective effects of Schwann cell derived neurotrophic factor (SDNF) on motoneurons of spinal anterior horn from spinal root avulsion induced cell death. Methods SDNF was applied at the lesion site following C 6,7 spinal root avulsion once a week. By the end of three weeks, the C 6,7 spinal region was dissected out for motoneuron count, morphological analysis and NOS expression histochemistry. Results In the control group, 68.6% motoneuron death of spinal cord occurred by 3 weeks following surgery, and the size of surviving motoneurons was significantly atrophy and NOS positive neurons increased. In the experimental group, the mortality was decreased by 35% as compared with that in the control group. No atrophy was found in the surviving motoneurons, and the expression of NOS was inhibited. Conclusion SDNF can prevent the motoneurons from death following spinal root avulsion. NO may play a role in the injury induced motoneuron death.
