乌拉坦对小鼠胚胎肝细胞发育分化的影响

摘    要:用伴刀豆凝集素(ConA)-辣根过氧化物酶(HRP)定位技术观察乌拉坦经胎盘对胚胎鼠及新生鼠肝细胞ConA受体分布变化的影响。结果显示:(1)13~15 d胚胎肝细胞ConA受体主要分布于细胞膜及核膜;17~19 d胚胎肝细胞胞膜ConA受体增多,核膜受体消失,胞浆出现ConA受体;3~14 d新生鼠ConA受体主要分布于胞浆,胞膜和核膜的ConA受体基本消失。(2)用乌拉坦后可减慢胞膜上ConA受体的消失,延迟胞浆ConA受体的出现,尤其对11~15 d胚胎作用最为明显。提示正常胚胎肝细胞ConA受体分布随肝细胞成熟呈规律性变化;乌拉坦可能在分子水平干扰了胚胎肝细胞的正常发育与分化。

Abstract:This article deals with the transplacental effects of Urethan on the distribution of ConA receptors in the embryo liver cells and the newborn liver cells of mice with labelling technique of Concanavalin-Horseradish Peroxidase (ConA-HRP). The results indicated as follows; (1)The ConA receptors were mainly found on nuclear and cell membrane of embryo liver of mice in 13-15 days, but the ConA receptors on liver cell membrane further increased by 17-19 days, with the appearence of cytoplasmic ConA receptors and the disappearance of nuclear membrane ConA receptors. Both the cell and the nuclear membrane receptors of ConA became negative on 3-14 days after the mice were born, while the plasma exhibted obvious ConA binding. (2)Urethan could delay the disappearence of cell membrane ConA receptors and the appearance of cytoplamic ConA receptors, especially when urethan was given during 11-15 days, which snggests that distribution of ConA receptors in the liver cell of normal mice embryo showed changes regularly. Urethan might have effects on development and differentiation of the mice embryo liver cell at the molecular Jevel, and lectin as a molecular probe provide to be considerable useful in studying the development and the differentiation of embryo cell with a molecular biology basis.
