针刀松解法与电针对膝骨关节炎大鼠中枢啡肽类物质影响的比较研究

摘    要:目的:观察针刀松解法及电针对膝骨关节炎大鼠中枢β-内啡肽(β-EP)及八肽胆囊收缩素(CCK-8)的影响。方法:将60只SD大鼠随机分为空白组、模型组、电针组和针刀组。用4%木瓜蛋白酶溶液与半胱氨酸溶液注入大鼠左膝关节腔复制膝骨关节炎模型。针刀组和电针组分别给予针刀松解法和电针治疗,疗程3周。采用放射免疫法检测脊髓及中脑β-EP及CCK-8的含量。结果:模型组大鼠脊髓及中脑β-EP和CCK-8含量较空白组显著增加(P<0.01);在脊髓水平针刀组及电针组β-EP含量较模型组升高,以电针组明显(P<0.05);针刀组和电针组脊髓和中脑CCK-8含量较模型组显著降低(P<0.05,P<0.01);两组间β-EP及CCK-8含量无显著性差异(P>0.05)。结论:针刀松解法和电针均可调节膝骨关节炎大鼠脊髓和中脑β-EP和CCK-8的合成和释放。

Abstract:Objective: To observe the effects of acupotomy lysis and electro-acupuncture on central β-endorphin(β-EP) and cholecystokinin-octopeptide(CCK-8) in knee osteoarthritis rats.Methods: Sixty SD rats were divided randomly into control group,model group,electro-acupuncture(EA) group and acupotomy lysis(AL) group.Knee osteoarthritis model was established by injection of caroid and cysteine into the left knee cavity.AL group and EA group were respectively given acupotomy lysis and electro-acupuncture therapy for three weeks.The contents of β-EP and CCK-8 were detected in the rats' midbrain and spinal cord with radioimmunoassay(RIA).Results: Compared with the control group,the contents of β-EP and CCK-8 in spinal cord and midbrain were significantly increased in the model group(P0.01);compared with the model group,the content of β-EP in the spinal cord were strikingly increased in AL group and EA group,with a more significant increase in the EA group than in AL group(P0.05);compared with the model group,the contents of CCK-8 in the spinal cord and midbrain were decreased notably in AL and EA groups(P0.05,P0.01).There were no differences in the contents of β-EP and CCK-8 between AL and EA groups(P0.05).Conclusion: Acupotomy lysis and electro-acupuncture can regulate the synthesis and release of β-EP and CCK-8 in the spinal cord and midbrain in knee osteoarthritis rats.
