摘    要:观察了刀豆蛋白A(ConA)诱导小鼠急性肝损伤模型的荧光组织化学和超微病理改变。小鼠注射ConA后,经超微病理观察发现,肝损伤突出表现为肝细胞凋落;用生物素化的ConA注射于小鼠,切肝制成冰冻切片,再加入异硫氰荧光素(FITC)标记的亲合素可见大部分荧光分布于肝脏。结果表明,细胞凋落可能是ConA在体内造成肝细胞死亡的机制;ConA具有亲肝特性。

Abstract:The purpose of this study was to examine changes in fluorescent histochemistry and ultrastructural pathology on concanavalin A (Con A)-induced liver injury in mice. Hepatic injury was characterized by apoptosis of liver cells through observation of electron microscope after Con A challenge. Livers were removed and divided into frozen sections after intravenous administration of Con A biotinylated to mice.When fluorescein isothiocyanate (FITC)-labeled Avidin was administered in the sections, the major fluorescence was found within the liver. This suggests that apoptosis is a possible mechanism of hepatocyte cell death induced by Con A in vivo and the distribution of Con A is specific for the liver.
