摘    要:目的:从基因水平了解淋病奈瑟菌对青霉素类、四环素类、喹诺酮类3种抗菌药物的耐药状况。方法:采用聚合酶链反应(PCR)对常州地区的50株淋病奈瑟菌进行TEM-1基因、tetM基因检测并对gyrA基因的DNA进行测序分析。结果:50株淋病奈瑟菌中32株TEM-1基因阳性,24例tet基因阳性,经PCR扩增和DNA测序,gyrA基因全部存在突变,PPNG—TRNG—QRNG3种分子检测发现青霉素类、四环素类与喹诺酮类二重耐药分别占26%和10%,耐多种药物(MDR)菌株总数已达74%。结论:淋球菌对青霉素类、四环素类、喹诺酮类抗菌药物的耐药性已相当高,迫切需要寻找新的敏感抗菌药物。

Abstract:Objective: To test the drug resistance gene of Neisseria gonorrhoeae to penicillin, tetracyclines and quinolones. Methods: Fifty strains of Neisseria gonorrhoeae were used to detect TEM-1, tetM and gyrA gene by means of polymerase chain reaction (PCR). DNA sequencing was also performed to test the gyrA gene. Results: PCR results were showned that 32 out of 50 strains of Nneisseria gonorrhoeae were TEM-1 gene positive and 24 were tetM gene positive. The mutation of gyrA gene was found in all these 50 strains by PCR and DNA sequencing. PPNG-TRNG-QRNG test revealed that in all these strains 74% of them were resistant to two or three drugs simultaneously (penicillins, tetracyclines and quinolones). Conclusion: There is a high ratio of drug resistance to penicillins, tetracyclines and quinolones which have been commonly used for gonococcal infection in clinic. The urgent need is to develop new effective antimicrobials.
