摘    要: 目的通过扫描电镜观察白屈菜红碱(CHE)对变形链球菌(S.mutans)作用后的形态学影响,进一步揭示其抑制细菌黏
附的作用机制。方法选择变形链球菌ATCC25175 作为实验菌株。将1.0 cm *1.0 cm *0.02 cm 的盖玻片放入15 mL试管中灭
菌备用。将CHE 溶液处理组、空白对照组及基础对照组3 组溶液及菌液分别加入灭菌后的试管中,在37 益厌氧环境(80%
N2,20%CO2)下培养24 h,细菌在盖玻片上形成一层生物膜,取出盖玻片,扫描电镜观察。结果97.5 μg/mL 的CHE 溶液可明
显抑制S.mutans对玻璃表面的黏附作用及细菌之间的相互黏附作用,表现为细菌胞体间黏附明显稀疏,黏附层薄,部分区域无
黏附。高倍镜下细菌胞体多呈胶囊状,细菌分裂横壁不明显,两端不对称且形态不规则,分裂后的细菌链局部出现断裂现象。
结论CHE 溶液对S.mutans的黏附有显著的抑制作用,在龋病防治领域具有一定的应用前景。

Abstract:Objective To observe the effect of Chelerythrine (CHE)on morphologic change of Streptococus mutans(S.mutans)using
scanning electron microscope (SEM),and to further reveal its mechanism of inhibitory effect on the adherence of . Methods
ATCC 25175 was used in this study. The study was divided into three groupsChelerythrine solution treatment group
(experimental group),blank control group and basic control group.S.mutans was cultured at 37 ℃in anaerobic environment (80 %N2,20
%CO2)for 24 hours,biofilms of S.mutans were formed on 1.0 cm *1.0 cm *0.02 cm standard glass microscope slides in 15 mL tubes,and
microscope slides were observed by SEM. Results Chelerythrine at the concentration of 97.5 μg/mL has significant inhibitory effect on the
adherence of S.mutans. There were fewer cells in chains,fewer cell layers in the experimental group than that of the control group. The cells
in cleavage formed into irregular morphous,most bacteria appeared as capsule-shaped cells. No clear cross wall of daughter cells was found
after division,and typical long bacteria chains were lost. Conclusion Chelerythrine has significant inhibitory effect on the adhesive function
of S.mutans,and might be used in the area of dental caries prevention and treatment in the future.
