摘    要:中药药代动力学以及中药药效物质基础长期以来都是中药学研究的瓶颈问题。考虑到中药化学成分的多样性及其复杂性带来的困难,以及内源性功能小分子对人类代谢影响的重要性,本文提出了整合药代动力学和代谢组学的方法。以代谢组为桥梁,从机体对中药产生的内源性小分子应答这一角度,通过同时监测 中药原型及被代谢成分(药代)和内源性小分子代谢物(代谢组)的变化,将二者通过多变量统计分析技术进行关联。这一方法合理简化了中药和人体这两个复杂系统,可以全面探索彼此的交互效应,考察单个药物成分在 这两个系统中的作用和地位。

Abstract:Studies on the pharmacokinetics and the material basis of Traditional Chinese Medicine (TCM) have always been the bottleneck of TCM research. Considering the difficulties arised form the complexity and the diversity of the chemical ingredients of TCM and the importance of endogenous metabolites played on human metabolic pathways, this paper proposed a method that integrated metabolomics and pharmacokinetics for material basis study of multi-component TCM. With metabolomics as the bridge, from the angle of metabolic response of the body to TCM, this method simultaneously detected the constituents of TCM and the changes of metabolites, endogenous metabolites in the body, which were then connected by multivariate statiscal data mining techniques. This strategy greatly simplified the systems of human body and TCM, which promoted exploration of mutual interaction of the two systems and investigation on the function and role of TCM components in the systems.
