摘    要:目的:通过气相色谱-质谱联用技术对中药名方保元汤的抗缺氧作用及途径进行研究,寻找生物标记物,阐明代谢通路。方法:小鼠随机分为6组(正常组、模型组、对照组和保元汤低、中、高3 个剂量实验 组),每组8只。灌胃给药12天后,将小鼠放入密闭具塞广口瓶中,以死亡时间作为抗缺氧能力指标。收集血液 进行血常规分析和气相色谱质谱联用仪分析。数据通过软件分析寻找差异性代谢物,并对代谢通路进行分 析。结果:保元汤各剂量组抗缺氧死亡时间比较模型组均有明显延长,代谢组学分析中共找到25个内源性标 志物。结论:保元汤具有显著性抗缺氧作用,通过代谢组学分析找到4条代谢通路,包括①甘氨酸、丝氨酸和苏 氨酸代谢路径,②氨酰-tRNA合成通路,③缬氨酸、亮氨酸和异亮氨酸代谢路径,④β-丙氨酸代谢路径。

Abstract:This paper was aimed toindentify metabolic biomarkers and metabolic pathways underlie the anti-anoxia effect of traditional Chinese medicine Baoyuan Decoction by gas chromatography-mass spectrometry (GC-MS).Mice were randomly divided into 6 groups (normal group, model group, control group, and low, middle, and high dose of three experimental groups), with 8 mice in each group. After 12 days of intragastric administration, the mice were placed in closed stoppered jars to moniter time. The time of death was used as an indicator of anti-anoxia effect, and the serum data was extracted by GC- MS. The data was analyzed by software to finding differential metabolites and analyzing metabolic pathways. The duration of anti-anoxia effect was significantly prolonged in each dose of Baoyuan Decoction, and 25 metabolic biomarkers were found in the metabolomic analysis. Conclusion: Baoyuan Decoction has significant anti-hypoxic effect, and 4 metabolic pathways were found through metabolomic analysis, including Glycine, serine and threonine metabolism. Aminoacyl- tRNA biosynthesis. Valine, leucine and isoleucine biosynthesis 2, beta- Alanine metabolism.
