阿尔茨海默病APPswe/PS1ΔE9双转基因小鼠脑区中老年斑的分布

摘    要:目的:观察老年斑在不同月龄的APPswe/PS1ΔE9双转基因小鼠脑区中的分布。方法:将雄性的杂合子小鼠与C57BL/6J雌鼠交配,所生的小鼠提取鼠尾DNA,PCR扩增目的基因。取鉴定为阳性的2.5、4.5、8月龄的杂和子小鼠各3只,进行Thioflavine S和Aβ免疫组织化学染色,观察老年斑的沉积。结果:共生产小鼠F1代39只,鉴定为阳性的有17只,阳性率为43.6%。组织学染色发现2.5月龄小鼠的脑区未见老年斑沉积,而4.5月龄及8月龄小鼠的皮质及海马可见老年斑沉积,并且随着月龄的增加,老年斑的数目增多,面积增大。结论:APP-swe/PS1ΔE9双转基因小鼠在4月龄时已有老年斑的沉积,为研究AD提供了很好的载体。

Distribution of Senile Plaque in the Brain of APPswe/PS1ΔE9 Double Transgenic Mice Model
Abstract:Objective: To observe the distribution of senile plaque deposition in transgenic mouse brain.Methods: Male heterozygote mice were mated with female C57BL/6J mice.DNA of each pup was extracted from the tail of mice and amplified by PCR.The brain section of heterozygote mice identified by PCR and aged 2.5 month,4.5 month and 8 month were stained with thioflavine S and Aβ to study the deposition of senile plaque.Results: Seventeen APPswe/PS1ΔE9 double transgenic mice were identified as positive from 39 pups(positive rate,43.6%).Immunostaining found senile plaque was detected in 4 month and 8 month old transgenic mice brain.There was no senile plaque deposition in mice brain aged 2.5 month.The number and area of the plaques increased with age.Conclusion: Senile plaque forms in APPswe/PS1ΔE9 double transgenic mice brain as early as 4 month.This kind of double transgenic mice is an efficient animal model for the study of Alzheimer's disease.
