人眼球冰冻切片制作实验研究

摘    要:目的探索人眼球切片制作的实验方法。方法收集捐献的新鲜成人眼球,用改良的AGFD眼球固定液[1]和丙酮固定,明胶包埋,冰冻切片机切片,HE染色,正丁醇透明[2],中性树胶封片。结果该方法制作的人眼球切片较为理想,可清楚观察睫状突和睫状小带,对显示视网膜和视盘微细结构也较好。结论经改良的AGFD眼球固定液加丙酮固定,很大程度上保持了眼球原有的形状。用明胶处理和冰冻切片法切片可简化操作过程,有效防止眼球破碎。制片较为理想。

Abstract:Objective To develop an experimental method for frozen sections of human eyeballs. Methods Fresh eyeballs collected from adult donors, were fixed with improved AGFD eyeball fixing solution and acetone. Then embedding them in gelatin, frozen sections, HE staining, clearing with n-butanol, and mount- ing with neutral gum were performed in turn. Resuls This method produces ideal ciliary processes and ciliary zonule sections. At the same time, it is a good method to observe the fine structure of retina, optic disc and so on. Conclusions This method preserves tissue morphology as well. Gelatin embedding and frozen sections could be used to simplify the performance, and tissue shrinking and crushing were also efficiently prevent from.
