| 摘 要： | 【目的】建立高效液相色谱（high performance liquid chromatographv，HPLC）紫外检测法测定反式二羟基环氧苯并（a）芘（＋）-anti-benzoa）pyrene diol-epexide，BPDE]在人体内形成的BPDE—DNA加合物的含量，对其质量进行控制。【方法】采用Kromasil—C18柱（150mm×4．6mm，5μm）为分析柱，以甲醇-水-磷酸（80：20：0．1，v／v）为流动相，流速1．0ml／min紫外检测器检测，检测波长254nm。【结果】在4．016-803．2ng／ml的浓度范围内，BPDE峰面积与浓度呈良好的线性，相关系数r=0．9997；平均回收率均在85％～115％之间，RSD为1．32％-1．72％。【结论】本法准确、方便，可用于测定空气中苯并（a）芘在体内代谢产物BPDE～DNA加合物的含量。 |

| Abstract: | Objective] To detect the levels of BPDE-DNA adducts by HPLC methods. Methods] The column was Kromasil C18; the mobilephase was methanol: water: phosphoric acid (80: 20: 0.1, v/v); the detective wavelength was set at 254 nm. Results] The linear range was 4.016～803.2 ng/ml (r=0.9997). The average recovery was be-tween 85% and 115% and the relative standard deviation (RSD) was less than 5.0%. Conclusions] This method is accurate, convenient and it can be used for the detection of BPDE-DNA. |
