摘    要:目的:观测择时服用人参皂甙对肝癌小鼠抗氧化能力及免疫功能方面的影响。方法:采用小鼠H22肝癌细胞实体瘤模型,分别于卯、酉时用药,检测肿瘤内总抗氧化能力、谷胱甘肽、超氧化物歧化酶(SOD)、血清IL-2、TNFα、VEGF、外周血白细胞数、脾脏及胸腺指数等指标。结果:①人参皂甙卯时用药肿瘤组织中的总抗氧化能力显著降低,SOD浓度显著升高,有显著性差异。②与模型组相比,人参皂甙可降低肝癌小鼠的血清IL-2、TNF-α和VEGF的含量,有显著性差异(P<0.05),但卯时组与酉时组之间无显著性差异(P>0.05)。③人参皂甙可使外周白细胞数升高,脾脏重量增加,有显著性差异(P<0.05),与用药时间无相关性(P>0.05),对胸腺重量无明显增重作用(P>0.05)。结论:人参皂甙可降低H22肝癌小鼠肿瘤组织的总抗氧化能力,与用药时间不同有相关趋势,并抑制机体的体液免疫功能,增强细胞免疫功能,与时间无相关性。

Abstract:Objective:To investigate the effects of time-selected administering ginsenoside on the mechanism of antioxidation and immunity in mince bering H22 hepatic cancer cells.Methods:After preparation of H22 hepatic cancer mice models,the animals were given ginsenoside at separate time,i.e.,maoshi(5 a.m.to 7 a.m.) and youshi(5 p.m.to 7 p.m.) and detected for the total antioxidant capacity and levels of glutathione(GSH),superoxide dismutase(SOD),IL-2,TNF-α and VEGF,white cell count,spleen index and thymus index.Results:Total antioxidant capacity in the tumor cells was significantly reduced,whereas SOD level was markedly increased when the ginsenoside taken at maoshi(P<0.05).Although ginsenoside greatly reduced the levels of IL-2,TNF-α and VEGF in blood serum as compared with the models(P<0.05),the two groups showed no significant difference regarding medication at maoshi and youshi(P>0.05).In addition,ginsenoside intake caused rise of white cell count and increased weight of spleen(P<0.05).The findings showed no relation to the time of medication and less effect on the weight increase of thymus gland(P>0.05).Conclusion:Ginsenoside may reduce the antioxidant capacity of tumor cells in H22 hepatic cancer mice models,inhibit humoral immunity but enhance cellular immune function,yet,there is no causative effect on experimental animals with the time of intervention.
