白细胞介素Ⅰ受体拮抗剂和抗白细胞介素6抗体对NZB/WF_1小鼠狼疮性肾炎的影响

摘    要:观察白细胞介素Ⅰ受体拮抗剂(IL-1ra)和抗白细胞介素6抗体(抗IL-6)对9月龄狼疮倾向NZB/WF1小鼠肾脏的形态学表现:IL-1ra组为轻度局灶性系膜增生性肾小球肾炎,病变单一;抗IL-6组呈中度弥漫性系膜增生性肾小球肾炎;盐水对照组为重度弥漫性系膜增生性肾小球肾炎伴明显病变多样性改变。电镜下3组肾小球内均可见多处电子致密物沉积,但IL-1ra组小且少,盐水组则大且多。结果提示,IL-1ra有明显减轻狼疮倾向NZB/WF1小鼠肾小球病理损害作用,从而延缓肾小球硬化,改善肾功能。抗IL-6组亦对小鼠肾小球病变有改善作用。

Abstract:The effects of interleukin-1 receptor antagonist (IL-1ra) and anti-interleukin-6 antibody (an-ti-IL-6 ) on renal morphological manifestation of lupus-like NZB/WF1 mice were observed. The resultsshowed that the lesion in IL-1ra group was focal mesenteric hyperplastic glomerular nephritis. The pathologicchange was single. In anti-IL-6 group, moderate disseminated mesenteric hyperplastic glomerular nephritiswas observed, while in the control group, the pathologic manifestation was severe disseminated mesenterichyperplastic glomerular nephritis with obvious multiple-change lesion. The electron microscope showed that,in all the three groups, the multiple electro-dense deposition was observed in the glomerulus. But the lesionin IL-1ra group was small and in less amount, while in the control group it was big and in larger amount.The results suggest that IL-1ra can obviously alleviate the glomerular pathologic damage in lupus-like NZB/WF1 mice, which might delay the sclerosis of glomerulus and improve the renal function. Anti-IL-6 also hascertain improving effects on the murine glomerular lesion.
