聚合酶链反应对脑脊液标本中结核杆菌DNA重复序列的检测

采用聚台酶链反应（ＰＣＲ）对３４例结核性脑膜炎（结脑）患者脑脊液（ＣＳＦ）进行检测，其中５份结核杆菌培养阳性的标本，ＰＣＲ检测均阳性；２９例ＣＳＦ结核杆菌培养阴性的标本，１９例ＰＣＲ检测阳性，总的阳性率为７０．５％。而作为对照的２０例非结脑患者的ＣＳＦ标本则无阳性。利用育法微量结核杆菌与对照菌测试，可以反证本方法的准确性，其符合率为１００％，提示本方法可以用于临床标本中结核杆菌ＤＮＡ的快速检测，不失为一快速，灵敏而又特异的诊断手段。关键词

DETECTION OF MYCOBACTERIUM TUBERCULOSIS DNA IN CEREBROSPINAL FLUID OF THE PATIENTS WITH TUBERCULOUS MENINGITIS USING PCR

n this study,we used the PCR protocol tO detect the repetitive DNA sequences ofMycobacterium tuberculosis in cerebrospinal fluid samples of patients with tuberculousmeningitis,the amplification protocol gave positive results in 5 of 5 cases which werepositive by culture as well as in l9 of 29 cases which were nagative by culture(overdallsensitivity 70.5%).The control san1ples were obtained from the patients with cryptococcusmeningitis,pyogenic meningitis or brain tumor,of the 20 cases not a sigle one waspositive. We conclude that the sensitivity of PCR in the detection of Mycobacteriumtuberculosis DNA in clinical samples is superior to that of standard culture technique.
