妊娠高血压疾病患者胎盘组织中细胞凋亡调控基因bax和bcl-2蛋白的表达及意义

摘    要:目的研究胎盘组织中细胞凋亡调控基因bax和bcl-2蛋白的表达与妊娠高血压疾病发病关系。方法采用免疫组织化学方法(SP),对10例正常晚孕妇和30例妊娠高血压的胎盘组织中bax和bcl-2蛋白的表达水平检测。结果bax在正常和轻度子痫前期、轻度子痫前期和重度子痫前期、重度子痫前期和子痫胎盘组织中呈增高趋势,但无显著性差异(P>0.05)。在正常和重度子痫前期、正常和子痫、轻度子痫前期和子痫胎盘组织中,bax表达明显增强(P<0.05,P<0.001,P<0.01)。bcl-2在正常、轻度子痫前期、重度子痫前期、子痫胎盘组织中表达呈增高趋势,但无显著性差异(P>0.05)。在正常和轻度子痫前期、轻度子痫前期和重度子痫前期、重度子痫前期和子痫胎盘组织中bax/bcl-2无显著性差异(P>0.05)。在正常和重度子痫前期、正常和子痫、轻度子痫前期和子痫胎盘组织中bax/bcl-2有显著性差异(P<0.05,P<0.01,P<0.05)。结论正常晚孕妇及轻度子痫前期中bax/bcl-2呈平衡趋势,重度子痫前期和子痫中呈失衡状态,即细胞凋亡调控基因蛋白bax和bcl-2蛋白是否平衡,将影响胎盘结构与功能,妊娠期高血压疾病的发生、发展与细胞凋亡失衡有关。

Abstract:Objective It is to study the expression of apoptotic controlling gene bax and bcl-2 proteins in placental tissue and the relation with onset of gestational hypertension.Methods The expression levels of bax and bcl-2 proteins in placental tissue of 10 cases of normal term pregnancy(NTP) and 30 cases of gestational hypertension were detected with immunohistochemical method(SP).Results The expression levels of bax in placental tissue between NTP and light eclampsism,light eclampsism and serious eclampsism,serious eclampsism and eclampsia were showed as heightening tendency,but the difference was not significant(P>0.05).The expression levels of bax in placental tissue between NTP and serious eclampsism,NTP and eclampsia,light eclampsism and eclampsia were obviously heightened(P<0.05,P<0.001,P<0.01).The expression levels of bcl-2 in placental tissue of NTP,light eclampsism,serious eclampsism and eclampsia were showed as heightening tendency,but the difference was not significant(P>0.05).There were no significant difference on bax/bcl-2 in placental tissue between NTP and light eclampsism,light eclampsism and serious eclampsism,serious eclampsism and eclampsia(P>0.05).There were significant difference on bax/bcl2 in placental tissue between NTP and serions eclampsism,NTP and eclampsia,light eclampsism and eclampsia(P<0.05,P<0.01,P<0.05).Conclusion The bax/bcl-2 is balance in NTP and light eclampsism,while the balance is broken in serious eclampsism and eclampsia.Apoptotic controlling gene bax and bcl-2 proteins is balance or not will affect the structure and the function of placenta.The occurrence and development of gestational hypertension have relation with apoptosis disequilibrium.
