小刺猴头菌丝体多糖的提纯及免疫与抗肿瘤活性的研究

摘    要:目的研究小刺猴头深层培养菌丝体多糖的免疫与抗肿瘤活性。方法从小刺猴头菌丝体中提出水溶性粗多糖,DEAE-cellulose及Sephadex G200柱色谱进一步纯化得到纯化的小刺猴头菌丝体多糖(Hericium caput-medusae polysaccharides A,HPA),气相色谱测定其单糖组成,药理试验考察其对S-180肉瘤、肝癌、艾氏腹水癌瘤株的抑制作用及对免疫功能的影响。结果 HPA为均一多糖,相对分子质量为2900,单糖组成为葡萄糖、半乳糖、甘露糖和木糖。对S-180肉瘤及艾氏腹水癌瘤株,HPA在20、40、80mg/kg时明显降低肿瘤的重量,呈现一定的肿瘤抑制率;能够显著增加荷瘤小鼠血清溶血素含量及血清凝集素水平。结论小刺猴头菌丝体多糖对提高免疫和抗肿瘤均有作用。

Abstract:Objective To study immunity and antineoplasmic activity of polysaccharides of Hericium caput-medusae mycelia of submerged-culture.Methods The water-soluble crude polysaccharides was isolated from the Hericium caput-medusae mycelia,and the Hericium caput-medusae polysaccharides A(HPA) was further purified by DEAE-cellulose column and Sephadex G200 column chromatography.The component of monosaccharide was studied by gas chromatography.The inhibitory action on tumor cell strain of S-180 sarcoma,hepatocarcinoma and Ehrlich's ascites carcinoma and the effect on immune function was studied by pharmaco logical test.Results HPA was a homogeneous polysaccharides,and its relative molecular weight was about 2 900.The component of monosaccharide was glucose,galactose,mannose and xylose.The level of 20,40,80mg/kg HPA could inhibit the growth of S-180 sarcoma and Ehrlich's ascites carcinoma:significant decreasing of the tumor weight and higher tumor inhibition rates.The content of erythrocytolysin in serum of tumor-bearing mice increased and the level of serum agglutinant boosted significantly.Conclusions HPA has an effect of antitumor and boosting immunity.
